Armand Rousseau inherited land in Gevrey-Chambertin through his 1909 marriage and expanded the domaine through the 1910s and 1920s with plots in Charmes-Chambertin, Clos de la Roche and Chambertin. On the advice of wine writer Raymond Baudoin, the estate began bottling and selling its own wine in the 1930s instead of selling grapes to négociants, and it was among the first Burgundy domaines to sell directly to American importers after the end of Prohibition. Further Chambertin-area plots followed, including Mazis-Chambertin in 1937, Mazoyères-Chambertin in 1940 and Clos Saint-Jacques in 1954. Armand Rousseau died in 1959 and his son Charles took over as sole winemaker.

Charles Rousseau ran the domaine for more than two decades and built export markets across Europe, North America, Australia, New Zealand and Asia. His son Eric Rousseau joined in 1982 and introduced green harvesting and mechanical soil work in place of chemical treatments, moving the vineyards toward organic viticulture while keeping traditional vinification methods such as ageing in oak barrels. Eric's daughter Cyrielle Rousseau, trained in geology and viticulture, joined the domaine in 2014. The property farms grand cru parcels including Chambertin, Chambertin-Clos de Bèze, Clos de la Roche, Charmes-Chambertin, Mazis-Chambertin and Clos des Ruchottes, alongside the premier cru Clos Saint-Jacques in Gevrey-Chambertin, entirely with pinot noir.
