Vine growing at Haut-Brion is documented from 1423. Jean de Pontac assembled the estate after his marriage in 1525 and built its early reputation; his descendant Arnaud III de Pontac continued the family's ownership from 1649. Samuel Pepys recorded the earliest known tasting note of the wine in 1663, and in 1787 Thomas Jefferson visited the estate and bought six cases for his Virginia cellar. Joseph de Fumel, who owned Haut-Brion at the time, was guillotined in 1794 during the French Revolution, and the property changed hands several times over the following century, including a period under Talleyrand from 1801 to 1804 and later the Larrieu family.

American financier Clarence Dillon bought Château Haut-Brion in 1935 and founded Domaine Clarence Dillon to run it. His granddaughter Joan Dillon, Duchess of Mouchy, led the company from 1975 to 2008 and oversaw its purchase of Château La Mission Haut-Brion in 1983. Her son, Prince Robert of Luxembourg, has been president of Domaine Clarence Dillon since 2008. The vineyard lies on gravel soil raised slightly above the surrounding land, planted mostly to Merlot and Cabernet Sauvignon for the red wine, with a small production of white wine from Sémillon and Sauvignon Blanc.
